What Is PT-141?

PT-141 (Bremelanotide) is a synthetic cyclic heptapeptide with the structure Ac-Nle-cyclo[Asp-His-D-Phe-Arg-Trp-Lys]-OH. It was developed as a metabolically stabilized analog of the naturally occurring alpha-melanocyte-stimulating hormone (alpha-MSH), incorporating a non-natural amino acid and a lactam cyclization to improve stability and receptor selectivity.

In experimental settings, PT-141 is used as an agonist tool for the melanocortin receptor family. While its parent peptide is a non-selective melanocortin ligand, structural modification shifts activity toward the MC3R and MC4R subtypes implicated in central nervous system signaling, while reducing activity at MC1R associated with melanogenesis.
